Garland explained that his adaptation was based solely on the first novel in the trilogy: “When I began working on Annihilation, only the first book was available. I knew it was intended to be a trilogy, but I focused on the manuscript I had.” He described his approach as creating “a memory of the book” rather than a direct screen adaptation, aiming to capture the “dreamlike quality” and tone he experienced while reading VanderMeer’s work. Instead of sticking closely to the original narrative, Garland took the story in a unique direction, with VanderMeer’s approval. He chose not to read the other two books once they were published, fearing it would prompt him to revise his script. When he later heard about elements from the sequels, he was surprised by some similarities to his own writing.

In Annihilation, Lena, a cellular biology professor and former U.S. Army soldier, is interrogated after being the sole survivor of an expedition into the Shimmer, a mysterious zone that emerged from a meteor at a lighthouse in Florida. Three years after its arrival, many teams have entered the Shimmer, but only Lena’s husband, Kane, returns, unable to explain his experience. As Kane’s health deteriorates, Lena joins a new scientific expedition led by psychologist Dr. Ventress, accompanied by a geomorphologist, a paramedic, and a physicist. Inside the Shimmer, communication fails, and the team encounters mutated flora and fauna. They discover disturbing evidence of Kane’s previous expedition, including a soldier transformed into a lichen colony. Tensions rise as paranoia spreads, leading to violent confrontations. A mutant bear attacks, further fracturing the group.
Lena and Ventress press on to the lighthouse, the Shimmer’s epicenter, where Lena finds a videotape of Kane revealing a shocking act before his death. Ventress disintegrates into a shimmering entity that mimics Lena. In a desperate move, Lena tricks the creature into igniting a grenade, causing the Shimmer to dissipate. Afterward, Lena learns Kane’s condition has stabilized. When she confronts his doppelgänger, their encounter ends ambiguously as they embrace, their irises shimmering, leaving the nature of their identities in question.
